Hematological and biochemical responses of pirarucu (Arapaima gigas, Arapaimidae) fed with diets containing a glucomannan product derived from yeast and algae Acta Amazonica
HOSHINO,Maria Danielle Figueiredo Guimarães; MARINHO,Renata das Graças Barbosa; PEREIRA,Diogle Farias; YOSHIOKA,Eliane Tie Oba; TAVARES-DIAS,Marcos; OZORIO,Rodrigo Otavio de Almeida; RODRIGUEZ,Anselmo Fortunato Ruiz; RIBEIRO,Ricardo Amaral; FARIA,Fernando Sérgio Escócio Drummond Viana de.
ABSTRACT The hematological and biochemical responses of pirarucu fingerlings (Arapaima gigas) fed with diets containing different concentrations of a glucomannan product derived from yeast and algae were evaluated in order to ascertain the effect of these diets on fish physiology. Four treatments were conducted, with three replications, with 12 fish in each tank. The product evaluated (MycosorbA+(r)) was incorporated into the commercial diet, at four concentrations: 0, 1, 2 and 4 g.kg-1, called M0%, M0.1%, M0.2% and M0.4%, respectively. After 45 days of feeding, blood samples from six fish in each replicate were collected to perform the analyses. Their weight and length were determined to calculate the condition factor and weight gain, but no differences...

Influence of the pH of glutaraldehyde and the use of dextran aldehyde on the preparation of cross-linked enzyme aggregates (CLEAs) of lipase from Burkholderia cepacia Electron. J. Biotechnol.
Caballero Valdés,Eduardo; Wilson Soto,Lorena; Aroca Arcaya,Germán.
The preparation of cross-linked enzyme aggregates (CLEAs) of lipase has been a challenge due the low amount of lysine residues that lipases have on their surface. The results show that CLEAs prepared using dextran aldehyde (100-200KDa) have a higher hydrolysis activity and particle size (activities between 3186 ± 21 U/g of CLEA and 4800 ± 30 U/g of CLEA and particle sizes between 52.6 ± 18.7 µm and 126.2 ± 53.5 µm) than CLEAs prepared with glutaraldehyde (0.1 KDa) (activities between 894 ± 16 U/g of CLEA and 2874 ± 20 U/g of CLEA and particle sizes between 21.2 ± 5.1 µm and 83.4 ± 24.9 µm); Thermal stability assays of bioctalysts at 60ºC at pH 7.0 using phosphate buffer 25 mM showed that CLEAs prepared with dextran aldehyde have lower residual...

Oxidation of the albumin thiol to sulfenic acid and its implications in the intravascular compartment BJMBR
Turell,L.; Carballal,S.; Botti,H.; Radi,R.; Alvarez,B..
Human serum albumin (HSA) is the most abundant protein in the intravascular compartment. It possesses a single thiol, Cys34, which constitutes ~80% of the total thiols in plasma. This thiol is able to scavenge plasma oxidants. A central intermediate in this potential antioxidant activity of human serum albumin is sulfenic acid (HSA-SOH). Work from our laboratories has demonstrated the formation of a relatively stable sulfenic acid in albumin through complementary spectrophotometric and mass spectrometric approaches. Recently, we have been able to obtain quantitative data that allowed us to measure the rate constants of sulfenic acid reactions with molecules of analytical and biological interest. Kinetic considerations led us to conclude that the most...

Quenching of the intrinsic fluorescence of bovine serum albumin by chlorpromazine and hemin BJMBR
Silva,D.; Cortez,C.M.; Louro,S.R.W..
The binding of chlorpromazine (CPZ) and hemin to bovine serum albumin was studied by the fluorescence quenching technique. CPZ is a widely used anti-psychotic drug that interacts with blood components, influences bioavailability, and affects function of several biomolecules. Hemin is an important ferric residue of hemoglobin that binds within the hydrophobic region of albumin with high specificity. Quenching of the intrinsic fluorescence of bovine serum albumin (BSA) was observed by selectively exciting tryptophan residues at 290 nm. Emission spectra were recorded in the range from 300 to 450 nm for each quencher addition. Stern-Volmer graphs were plotted, and the quenching constant estimated for BSA solution titrated with hemin at 25ºC was 1.44 (± 0.05) x...

Radiation-induced Changes in the Electrophoretic Profile of Serum Albumin BABT
Lima,Celso Vieira; Campos,Tarcisio Passos Ribeiro.
ABSTRACT Albumin protein profiles were investigated in electrophoresis system in relation to the whole body exposition to the radiation. Two groups of rats Wistar were set up as the control (CG) and the irradiated one (IG). The IG was exposed to Co-60 at a dose of 5 Gy. After a 72-hour exposition, 300 μL of blood was collected in the inferior vena cava, renal, jugular, hepatic, and pulmonary veins and the serum separated. The albumin protein was identified by vertical electrophoresis in acrylamide Commassi blue or silver stained. The calibration procedure was applied to albumin samples with well-known concentrations. The mathematical correlation was developed involving electrophoretic parameters of band intensities and sizes from gel representation,...

Synergistic effect of microRNA and albumin-bound nanoparticles for inhibition of glioblastoma cancer cell proliferation BJPS
Shariatnasery,Maria; Irani,Shiva; Soleimani,Masoud; Goodarzi,Navid; Dinarvand,Rassoul.
The functional significance of upregulation miR-34a in combination with albumin-bound paclitaxel nanoparticles in U251 glioblastoma cell line has been evaluated. The MTT assay determined that miR-34a and albumin-bound paclitaxel nanoparticles can reduce cell viability, but the combination of both factors has a stronger effect on cell viability. The application of qRT-PCR has demonstrated that the transduction of miR-34a could lead to exogenous upregulation of miR-34a level and downregulation of SURVIVIN. Moreover, treatment of U251 cells with miR-34a and nanoparticles together considerably inhibit SURVIVIN expression compared to miR-34a and nanoparticles alone. Flow cytometry showed that upon miR-34a overexpression cell cycle arrested in G1 phase, while...

Using egg albumin foam to extinguish fires BABT
Alsaati,Hytham A.; Ditzler,Danielle; Burapatana,Vorakan; Tanner,Robert D..
Oil, coal and chemical fires are often difficult to put out using water. In certain hydrocarbon fires, protein foam can extinguish fires better than water by keeping air (oxygen) away from the flames and by ''blowing'' the flame away from its fuel source. Egg albumin is a relatively inexpensive protein and is representative of foaming proteins, which are candidates for use as fire suppression agents. This paper begins to deal with the effect of the foam bulk pH, foam protein concentration and generating air flow rate into the foam on the fire extinguishing time in laboratory experiments. A Bunsen burner was used to generate a small, controlled laboratory fire within a plastic container, which represented a point source in a partially open room in the...
